Test of the Skin-Messaging and Barrier Repair Actions of a Topical Emollient Formulated with Provitamin B5

RCT to test skin moisturizing and barrier repair abilities of a new topical emollient with Provitamin B5 (Panthenol) (NTP-CE). The study indicated that the application of NTP-CE was effective and persistent in barrier repair and moisturizing skin.

· Evaluation results
In the study (N = 23), two skin areas (one treated with 0.5% sodium dodecyl sulfate (SDS) solution and the other untreated) were treated with NTP-CE for 3 weeks. Transepidermal water loss (TEWL), skin moisture, and intercellular lipid layer (ICLL) organization were measured regularly during the study. The mean AUC reduction in TEWL relative to baseline was more significant in the NTP-CE group compared to the control group (-168.36 vs. -123.38 g/m 2 /h, p = 0.023). The use of NTP-CE was also associated with a significant improvement in stratum corneum hydration and an increase in mean ICLL length relative to baseline (Day 22: 120.61 vs. 35.85 nm/1000 nm2, p < 0.001).

Reduction of Skin Inflammation and Pruritus by Moisturizing Spray Containing Provitamin B5

The purpose of this work was to correlate skin inflammation, skin barrier function and pruritus in an AEW mouse model, and to examine the antipruritic effects of β-glucan and provitamin B5 (panthenol) used together in a moisturizing spray on this model. According to this research, the impaired barrier function of skin is linked to pruritus and inflammation. Adding β-glucan and provitamin B5 to moisturizing skin care products can relieve itching caused by dry skin, improve impaired skin barrier function, and reduce local skin inflammation.
· Glucan and panthenol (G & P) spray formulation
It contains β-glucan and provitamin B5 (panthenol) as active constituents. β-Glucan is a water-soluble extracellular glucan produced by a novel strain of Agrobacterium tumefaciens ZX09. Water, glycereth-26, propylene glycol, caprylhydroxamic acid, phenoxyethanol and chlorphenesin are the constituents of the base spray.
· Evaluation results
The application of a spray containing β-glucan and panthenol significantly mitigated the negative effects of AEW treatment. That led to fewer scratching episodes over an hour, less transepidermal water loss, higher levels of filaggrin mRNA, and reduced mRNA expression of pro-inflammatory markers: NOD-like receptor family pyrin domain containing 3, apoptotic-associated speck-like protein with a CARD, caspase-1, interleukin-1β, tumor necrosis factor-α, cathepsin S, thymic stromal lymphopoietin, and nerve growth factor in lesions.
