Here, a supramolecular collagen nanoparticle was synthesized by the encapsulation of palmitoyl tripeptide-5 (93.18%) into lactoferrin and recombinant collagen with its efficacy for skin care formulations was explored.
· Efficacy of supramolecular collagen NPs
The supramolecular collagen NPs were highly stable and the transdermal efficacy went up by 69.9%, reaching the dermis. After 28 days of use, the water content of the stratum corneum increased by 10.51%, facial elasticity was improved by 8.15%, skin firmness was increased by 12.53%, and the facial melanin index decreased by 1.84%. The 28.29% decrease in eye bags and 37.611% decrease in nasolabial folds were achieved 14 days later. It was also noticed wrinkles length and area reduced by 10.22% and 21.57% respectively and 34.41% and 27.92% respectively. The supramolecular collagen NPs are skin-filler, whitening, anti-wrinkle, and firming.
· Preparation of supramolecular collagen NPs
They supramolecular collagen NPs were made by an ordinary emulsion making procedure. The oil phase was created by heating, in an oil bath at 60°C, squalane (3.0 wt %), hydrogenated lecithin (0.5 wt %), PEG-40 hydrogenated castor oil (3.0 wt %) and vitamin E acetate (0.5 wt %). Palmitoyl tripeptide-5 (0.1%) was added to this solution and beaten together until well blended. The water phase was glycerol (5 wt %), 1,2-pentanediol (3.0 wt %), lactoferrin (0.2 wt %), recombinant human collagen (0.5 wt %), and treated water (84.2 wt %). Under shear the oil phase was incorporated gradually into the aqueous phase and mixed. These final supramolecular collagen NPs were created by homogenising the suspension six times at 10,000 psi with a high-pressure microfluidizer.

This work evaluated the efficacy of hyaluronic acid microneedle patches containing bioactive ingredients, which were formulated with arginine/lysine peptides, acetyl octapeptide-3, palmitoyl tripeptide-5, adenosine, and seaweed extract as active ingredients.
HA-MNs patch formulation
The HA-MNs patch was created using DAB technology, where each droplet was air-cured to form microneedles. The ingredients of the test product include arginine/lysine peptide (S-mu-conotoxin CnIII acetate), palmitoyl tripeptide-5, acetyl octapeptide-3, Undaria pinnatifida extract, Corallina officinalis extract, adenosine, and hyaluronic acid.
Evaluation methods and results
In a 12-week clinical trial conducted at a single center, HA-MNs patches were applied to the outer corners of both eyes and specific areas on the volar forearm of healthy subjects with aging skin. The product was well-tolerated, with no participants reporting any primary or cumulative skin reactions. Evaluations of measurable skin properties revealed significant effectiveness: fine lines and wrinkles decreased by 25.8%, skin hydration improved by 15.4%, and dermal skin density and thickness increased by 14.2% and 12.9%, respectively.
