Panthenol, the biologically active alcohol analogue of pantothenic acid is a pro-vitamin of the B-complex group that is a normal constituent of skin and hair. This study measured skin moisturizing capacity of formulas that contained various levels of panthenol. The results showed that the panthenol-based formulations increased skin moisture and had a significant effect on the skin barrier function by reducing the TEWL value. By including 1.0% panthenol, the formulation increased its protective activity and preserved the integrity of skin.
Evaluation Methods and Results
· These formulations, with or without 0.5%, 1.0%, or 5.0% panthenol added, were used every day on the forearms of healthy participants. The skin moisture and transepidermal water loss (TEWL) profiles were analyzed before and after the application period of 15 and 30 days. After 30 days of application, the panthenol-containing formulations (1.0% and 5.0%) resulted in a significant reduction in TEWL.
· The formulations were also applied after washing the skin with sodium laureth sulfate (SLES) to evaluate the direct effect on TEWL and skin moisture. In skin cleansed with SLES, TEWL was significantly reduced two hours after application of the formulation containing panthenol compared with the control and vehicle.

To evaluate the efficacy of panthenol in cosmetic and pharmaceutical semi-solid formulations, this work added 5-13 wt% panthenol to o/w and w/o emulsions to investigate whether these formulations can increase moisturizing effects and enhance the skin's Barrier performance.
