A film composed of sodium alginate-magnesium aluminum silicate (SA-MAS) was created and tested for use as a film former for dispersions to control drug release from tablets, thanks to the interaction between MAS and SA to form a phase-separated microcomposite.
· Preparation of SA and SA-MAS films
SA and SA-MAS composite films were prepared using a casting/solvent evaporation method. SA (2 g) was dispersed in distilled water with a homogeniser for 5 minutes, diluted to a final volume of 200 ml, and kept overnight at room temperature. The dispersion was cast on to a plastic sheet (15 cm x 20 cm) and dried at 50°C. The films were peeled and put in a desiccator. To incorporate MAS into the films, pre-hydrated MAS (1 g, 2 g, or 3 g) was mixed with the SA dispersion using a homogenizer for 5 minutes, followed by the casting and drying process.
· Preparation of SA-MAS coated tablets
The tablet formulation consisted of 10% active ingredient (propranolol HCl (PPN) or acetaminophen (ACT)), 30% microcrystalline cellulose, 58.8% spray-dried lactose, 0.2% colloidal silicon dioxide, and 1% magnesium stearate. The tablets were coated with a 3% w/v low-viscosity SA (LV-SA) dispersion or a 2% w/v LV-SA and 2% w/v MAS dispersion. Coating conditions were as follows: tablet weight 900 mg, inlet air temperature 60-65°C, spray rate 4 ml/min, and spray pressure 0.28 MPa.

This study suggested that lower doses of aluminum magnesium silicate (AMS)-stabilized antimicrobials combined with immunostimulants may be used to prevent and treat drug-resistant infections.
· Mechanism of Action
Stabilization of ampicillin trihydrate, sulfadimethoxine, piperazine citrate, and chloroquine phosphate with pharmaceutical synthetic aluminum magnesium silicate and supportive treatment with immunostimulants can prolong their bioavailability, enhance the patient's immune response, and improve treatment efficacy, thereby clearing 95% or more of sensitive and resistant infections.
· Efficacy evaluation of AMS-stabilized antimicrobials
The recommended dosages of Piparazine, Ampicillin, and Chloroquine achieved clearances of only 82.94%, 80.68%, and 20% for infections caused by Helignosomoides bakeri, Salmonella gallinarum, and Plasmodium berghei, respectively. Administering lower doses (75%) of AMS-stabilized Piparazine and Ampicillin resulted in clearances of 96.82% and 97.84% for the respective infections. When the AMS-stabilized Chloroquine and AMS-stabilized Ampicillin were combined with immune stimulants, complete clearance of P. berghei infections was achieved, along with a clearance rate of 95.80% for Ampicillin-resistant Escherichia coli infections.
What is Magnesium Aluminum Silicate and what are its primary uses in cosmetic formulations?
Magnesium Aluminum Silicate (MAS) is a white, highly purified mineral derived from smectite clays. It is primarily used in cosmetic formulations as a stabilizing, suspending, and emulsifying agent. This mineral enhances the stability and texture of products by acting as a co-emulsifier, thickener, and stabilizer.
How does Magnesium Aluminum Silicate improve the performance of skincare products?
MAS improves the performance of skincare products by controlling viscosity and rheology, especially in creams. Its high gel strength to viscosity ratio allows it to act as a viscosity-controlling agent. MAS also enhances sensory properties, resulting in formulations that are stable and aesthetically pleasing.
What advantages does Magnesium Aluminum Silicate offer for formulations with acidic preparations?
Thanks to its high acid compatibility, Magnesium Aluminum Silicate is particularly useful in acidic preparations. This feature allows it to maintain the stability and consistency of formulations that contain acids without compromising their effectiveness or texture.
Can Magnesium Aluminum Silicate absorb flavors and fragrances?
Yes, smectite clays, from which MAS is derived, are tasteless and odorless in their natural state but possess the ability to absorb certain flavors and perfumes. This property helps in masking bitter, medicinal flavors and prolongs the fragrance of perfume in formulations, making them more palatable and enjoyable for consumers.
How does Magnesium Aluminum Silicate contribute to the stability of emulsions?
Magnesium Aluminum Silicate acts as a protective colloid by surrounding oil droplets and keeping them dispersed. Its colloidal structure prevents coalescing and stabilizes both oil-in-water and water-in-oil emulsions, even at low concentrations and high temperatures. This enhances the stability and longevity of emulsions in cosmetic formulations.
What role does Magnesium Aluminum Silicate play in viscosity control?
As a viscosity controlling agent, MAS provides flexibility in adjusting the thickness and texture of formulations. When combined with natural or synthetic organic gums like Xanthan Gum, Cellulosics, or Acrylic Polymers, it helps achieve specific performance requirements by developing an internal structure that supports fine particle suspension and enhances product spreadability and film-forming properties.
Is Magnesium Aluminum Silicate compatible with other ingredients?
Yes, MAS is highly compatible with a variety of ingredients, including surfactants and other stabilizers. Its ability to blend well with diverse components makes it a versatile ingredient in creating stable, effective, and high-quality cosmetic and skincare products.
