This work evaluated the hydrophobic peptide MTD 1067 as a cell penetrating peptide (CPP) to transport protein cargos of various sizes such as growth hormone-releasing hexapeptide-6 (GHRP-6) and other growth factors [des(1-3)IGF-I and PDGF-BB] via transdermal application. Of them, GHRP -6 is referred to in the cosmetics industry as hexapeptide-2 (HP-2) and is now employed as an active ingredient for skin whitening by using its melanogenesis inhibition role as an -melanocyte stimulating hormone (-MSH) antagonist.
· Synthesis of MTD 1067-bound GHRP-6
The peptides GHRP-6 and MTD-GHRP-6 were chemically synthesized using the SPPS-peptide synthesis method. The purity of GHRP-6 and MTD-GHRP-6 was 97.67% and 96.55%, respectively, and the molecular weights of the final synthetic peptides were 873.3 Da and 1898.7 Da, respectively.
· Enhanced Effects
Compared to the original cargo, MTD 1067-bound GHRP-6 exhibited the same or enhanced biological activity. Compared to the control group without MTD, the level of MTD-GHRP-6 detected in the dermis was 4.4 times higher, with no cytotoxicity exhibited. MTD 1067-bound GHRP-6 can be used to develop cosmetic and pharmaceutical functional macromolecules with enhanced transdermal permeability.

CD36, as one of the growth hormone releasing peptide 6 (GHRP-6, also known as hexapeptide-2) receptors, is present in large quantities in the granulation tissue of skin wounds. This work investigated the healing response in the presence of GHRP-6 as a CD36 agonist. Correlative experimental evidence supports the hypothesis that a GHRP-6/CD36-mediated anti-inflammatory and anti-fibrotic loop exists that appears to improve wound closure and aesthetics.
· Evaluation Methodology
Excisional full-thickness wounds (6 mmØ) were made on the back of Wistar rats and topically treated twice daily for 5 days. A universal model of rabbit ear hypertrophic scarring was implemented, with animals treated daily for 30 days. Treatment for both species was based on a sodium carboxymethylcellulose (CMC) jelly composition containing GHRP-6 400 μ g/mL.
· Experimental Results
Experiments in rats have shown that GHRP-6 pharmacodynamics involve attenuation of immune-inflammatory mediators and their effector cells, as well as a reduction in the expression of profibrotic cytokines. Importantly, in a hypertrophic scar rabbit model, GHRP-6 intervention significantly reduced the occurrence of hypertrophic scars by activating PPARγ and reducing the expression of profibrotic cytokines.
