A completely plant-based nanofiber dressing composed of cellulose acetate (CA) and soy protein hydrolysate (SPH) was developed using rotary jet spinning. The obtained CA/SPH nanofibers successfully mimicked the physicochemical properties of the natural skin extracellular matrix (ECM) and exhibited a high water retention capacity.
Preparation and properties of CA/SPH nanofibers
· CA and CA/SPH, with varying compositions and concentrations (wt/v%), were dissolved in hexafluoroisopropanol (HFIP) and stirred overnight. For comparison, PCL at 6 wt/v% was also dissolved in HFIP. After mixing, the solutions were delivered to a rotating reservoir via polyfluoroalkoxy alkane tubing at a rate of 2 mL per minute using an automatic syringe pump. Subsequently, the solutions (totaling 10 mL) were sprayed from the reservoir at 60,000 rpm for 5 minutes, which elongated the polymers into nanofibers while rapidly evaporating HFIP from the orifice (360 µm in diameter). The resulting nanofibers were left to dry overnight in a desiccator to ensure the complete removal of any residual solvent.
· In vitro studies showed that CA/SPH nanofibers enhanced fibroblast proliferation, migration, infiltration, and the expression of integrin β1. In vivo, CA/SPH scaffolds facilitate re-epithelialization, promoted epidermal thinning, and minimized scar formation and collagen anisotropy, similarly to other fibrous scaffolds.
