Laurocapram and its analogues act as absorption enhancers and are used to temporarily increase the permeability of membranes (e.g., skin and mucous membranes) to achieve an enhanced effect on most hydrophilic and lipophilic substances.
Mechanism of action and research on laurazepam formulations
· Laurocapram is known to work through various mechanisms, such as inserting its dodecyl group into the intercellular lipid bilayer, increasing the motion of lipid alkyl chains, and fluidizing the hydrophobic regions of the lamellar structure.
· Numerous studies have been conducted using formulations containing Laurocapram in different vehicles and drugs to assess its enhancing effects. The impact of Laurocapram has been examined through both in vitro studies, using diffusion cells or skin isolation and perfusion, as well as in vivo permeation studies employing techniques like tape-stripping, radioactive markers, drug recovery from the skin's surface, and evaluation of biological or pharmacological effects.
· Studies have shown that Laurocapram enhances the permeation of most drugs, whether they are lipophilic or hydrophilic. Sometimes, the co-application of Laurocapram can even enhance the biological or pharmacological effects of certain drugs. However, recent research has indicated that in some cases, the expected enhancement effect of Laurocapram may not occur. For example, in studies involving drugs like 5-(2-bromovinyl)-2-deoxyuridine and cidofovir, the application of Laurocapram did not lead to the anticipated result. Additionally, with morphine hydrochloride, while iontophoretic transport was not increased with Laurocapram, less electric current was required for transport.
