This work developed a self-nanoemulsifying drug delivery system (SNEDDS) based on calendula oil (CO) and modified with chitosan (CS), using Nonoxynol-10 as surfactant. The optimized formulation of nanoemulsion (NE) can deliver posaconazole (PSZ) for the treatment of gingivitis.
Selection of surfactants and preparation procedures of PSZ-CO-CS
· In terms of PSZ's solubility in surfactants, the findings revealed that the highest solubility was achieved in Nonoxynol-10 (25 mg/mL) while the lowest solubility was observed in Polyoxyl 20 cetostearyl ether (7.5 mg/mL). As a result, Nonoxynol-10 was chosen as the surfactant to be used in the further development of the NE formulation.
· The preparation procedure of PSZ-CO-CS involved two stages. The first step included creating a basic SNEDDS with a constant amount of Smix 1:2 mixed with 6%, 12%, and 18% CO. In the second step, PSZ was added to the SNEDDS in amounts of 10, 15, or 20 mg. The final NE was prepared by mixing the PSZ-loaded SNEDDS with an aqueous dispersion of CS. A preliminary study was done to determine the maximum amount of PSZ that the NE could carry. The loading efficiency for all formulations was found to be over 98%. The optimized formula had a loading efficiency of 100%.
