This work demonstrated that the use of high-frequency ultrasound and coupling gel containing skin whitening agents can effectively improve facial pigmentation by enhancing the transepidermal transport of the whitening agents. The whitening gel sample was a coupling gel containing 2% ascorbyl glucoside and 3.5% niacinamide as whitening active agents.
Experimental methods and results
· The impact of ultrasound on the absorption of skin-lightening agents into the stratum corneum was examined using a tape-stripping method on human forearms in vivo. A facial clinical trial involving 60 participants with hyperpigmentation was conducted in a paired design to evaluate the efficacy of the skin treatments.
· The participants were split into two groups, with each group receiving two different treatments on each facial cheek: skin-lightening gel with ultrasound versus no treatment, and skin-lightening gel with ultrasound versus skin-lightening gel treatment. Changes in facial hyperpigmentation were measured through computer analysis and visual grading of high-resolution digital images of the face, as well as through a subjective questionnaire.
· The results showed that ultrasound radiation increased the absorption of skin-lightening agents in the stratum corneum in a time-dependent manner. In the facial clinical trial, the combination of ultrasound radiation and skin-lightening gel led to a significant reduction in facial hyperpigmented spots compared to both no treatment and using the skin-lightening gel alone after 4 weeks.
